Is there a protocol needed for initiating a PhoxyPlate?
The pH sensor used in the PhoxyPlates requires a pre-soaking phase to be activated.
In general, the pH sensitive membrane soaks up a bit of buffer/water/media to allow for fast and sufficient proton diffusion between sample and sensor molecules. We recommend soaking at least for 1 hour, better 3 hours. As soaking liquid, we recommend directly using the media or buffer that you want to use your samples in later on.
Subsequent to the soaking phase, please perform the Autodetect centers, Autoexposure and One Point Adjustment OPA. If you have already performed Autoexposure and OPA at the beginning, please re-perform after soaking to adapt for potential signal changes upon activation of sensor membranes.
O2 sensors do not need a pre-soaking phase for activation. Nevertheless, please consider the temperature dependencies of the sensor signals prior to OPA and starting an experiment.
